Strain developmentDeveloped by Yoko Yamagata and Yuchio Yanagawa, National Institute for Physiological Sciences in 2003. Derived from TT2 ES cell line (B6CBAF1 derived). The PGK-neo cassette was removed by transient expression of cre recombinase in ES cells. C57BL/6 background (RBRC05821), ICR mixed background (RBRC05636).
Strain descriptionCamk2a/K42R knock-in mice. Ca<2+>/calmodulin-dependent protein kinase IIalpha (CaMKIIalpha/ Camk2a) is an enzyme that adds phosphates to a variety of protein substrates to modify their functions. CaMKIIalpha is especially enriched in the hippocampus, and is believed to be an essential mediator of activity-dependent synaptic plasticity and memory functions. We generated a novel kinase-dead knock-in mouse of the CaMKIIalpha gene that completely and exclusively lacks its kinase activity by introducing a point mutation (K42R) that prevents ATP binding necessary for phosphorylation reaction. Both heterozygous and homozygous mice are hyperactive, but otherwise indistinguishable from wild-type mice. They are fertile and without any obvious neurological symptoms, except for sporadic observation of mild limbic seizure. Homozygous mice are severely impaired with hippocampal synaptic plasticity and hippocampus-dependent memory formation. Suitable for studies of the mechanisms of LTP and learning and memory, and as a memory-deficit animal model.
