Strain development | Developed by Haruhiko Koseki, RIKEN in 2011. M1 ((129 x C57BL/6)F1) ES cells were used. This strain was backcrossed to C57BL/6J. |
Strain description | Tet3 gene floxed mice. Gene targeting for the Tet3 allele was done by flanking exons 8 and 9 with Cre-loxP sequences. Conditional deletion of Tet3 (exons 8 and 9) can be achieved by crossing the mice with specific cre deleters such as CreERT2. The TET3 protein is a dioxygenase that catalyzes the conversion of 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC) and further oxidized cytosine (5fC and 5caC), and plays a key role in active DNA demethylation. Gdf9-Cre-driven Tet3 deletion in early oocytes resulted in a complete absence of Tet3 messenger RNA in Tet3mat-MII oocytes and Tet3 protein in zygotes (Amouroux et al., 2016). This results in a lack of 5hmC formation in the paternal pronucleus following fertilization in zygotes. This conditional allele would be useful to study the roles of Tet3 in DNA de-methylation by combining with tissue specific Cre deleters. |
