DDX4 protein, encoded by Ddx4 gene, is an ATP-dependent RNA helicase belonging to DEAD (Asp-Glu-Ala-Asp) box protein family. This protein is also known as VASA or MVH, and is highly conserved in germline cells of various species. It has been reported that Ddx4 expression in mice is found in primordial germ cells from E10.5 to E11.5 in both males and females, and its expression continues throughout life. The Ddx4-CreERT2 KI mouse (RBRC11094) is a Cre driver mouse that expresses tamoxifen-induced Cre recombinase under the control of endogenous Ddx4 promoter. Both mating with a Cre-reporter R26GRR strain (RBRC04874) and tamoxifen administration have been shown to induce complete Cre-loxP recombination in male germ cells (sperm stem cells) and female germ cells (oocytes in primordial follicles). Thus, Ddx4-CreERT2 KI mouse is a useful tool for analyzing mechanisms of female oogenesis and male spermatogenesis, and this line is expected to be used to further elucidate molecular mechanisms of germ cell development.
