Concentrated fluorescent activity of AktPH-GFP at the leading edge (Arrow Heads) during chemotactic peptide-stimulated neutrophil migration was visualized by confocal fluorescence microscopy.

 

  PI3Ks (phosphoinositide 3-kinases) regulate many critical cellular responses by producing PI(3,4,5)P3 (phosphatidylinositol 3,4,5-trisphosphate). The EGFP-Akt transgenic mice express a fluorescent bioprobe for PI(3,4,5)P3/PI(3,4)P2 (phosphatidylinositol 3,4-bisphosphate) which is the pleckstrin homology domain of Akt (AktPH) and EGFP fusion protein. The AktPH-GFP fluorescence shows defused localization in cytoplasm, however the fluorescence is concentrated at the leading edge during fMet-Leu-Phe (N-formylmethionyl-leucylphenylalanine; chemotactic peptide)-stimulated neutrophil migration. These transgenic mice are useful for studying PI(3,4,5)P3 metabolism in physiological and pathological situations by crossing your mutant mice.
