GABAergic neurons play an important role in the regulation and stabilization of network activities and are essential for a number of brain functions including cognition and emotion. Because GABAergic neurons are primarily scattered throughout the central nervous system, it is difficult to identify these cells in living brain preparations. A specific marker for GABAergic neurons is the enzyme glutamate decarboxylase 67 (GAD67). GAD67-GFP knock-in mice were generated using a gene targeting method via homologous recombination in embryonic stem cells [1, 2]. GFP fluorescence was observed specifically in the majority of GABAergic neurons in these knock-in mice, and these neurons could be detected easily by fluorescent microscopy without immunostaining. GAD67-GFP knock-in mice have facilitated the study of both the electrophysiological and anatomical profiles of GABAergic neurons [3-5].
