E14.5 (top) and E20.5 (bottom) embryo of PDGFRα-GFP/CreERT2 mice crossed with ROSA26-Tomato (left) or ROSA26-LacZ (right) reporter mice. Cre activity was induced by intraperitoneal injection of 4-hydroxytamoxifen in pregnant female mice at E9.5.

Mesoderm and its descendant cells arerequired for organogenesis, wound repair, and disease pathogenesis. It can be classified into cells positive for vascular endothelial growth factor receptor 2 (VEGFR2 or FLK1) or platelet-derived growth factor receptor α (PDGFRα), representing lateral and paraxial mesoderm, respectively. Era and colleagues generated knock-in mice carrying GFP-Cre-ERT2 at the start codon of Pdgfra [1]. They confirmed Cre activity in mesoderm cells at gastrulation and the mesenchyme of E9.5. Neural crest cells expressing PDGFRα was also confirmed to contribute to the facial mesenchyme. PDGFRα-GFP/CreERT2 mouse strain is a useful tool to investigate novel role of PDGFRα-expressing cells and their descendant cells in vivo.
